Overview

gp160MN + gp160LAI-2 is a bivalent recombinant glycoprotein vaccine developed for the prevention and treatment of HIV-1 infection. The product consists of the full-length envelope precursor protein, gp160, derived from two distinct HIV-1 Clade B isolates: the MN strain and the LAI (specifically the LAI-2/BRU) strain. The MN strain was selected for its high prevalence and relevance to circulating variants in North America and Europe, while the LAI strain represents one of the earliest and most extensively characterized HIV isolates. Produced using recombinant DNA technology, typically in a baculovirus expression system, the vaccine was designed to induce neutralizing antibodies and prime cellular immune responses by presenting the structural motifs of the viral envelope before its natural cleavage into gp120 and gp41 subunits. Clinical evaluation, conducted primarily in the 1990s through the AIDS Vaccine Evaluation Group (AVEG), explored the vaccine's potential as both a prophylactic agent in seronegative individuals and an immunotherapeutic agent in HIV-infected patients. However, like most first-generation envelope protein-based vaccines, it failed to demonstrate significant protection against infection or durable control of viral load, leading to the cessation of its development.
