GP350 CAR-T is an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target the gp350 glycoprotein of the Epstein-Barr virus (EBV). Developed by Zhimin Zhai and colleagues, the therapy is primarily intended for the treatment of EBV-associated lymphoid neoplasms, including Burkitt lymphoma and other EBV-positive malignancies. The CAR construct features single-chain variable fragments (scFvs) derived from gp350-binding, neutralizing monoclonal antibodies, which are fused to a signaling backbone containing 4-1BB (CD137) costimulatory and CD3ζ activation domains. By targeting gp350—a glycoprotein expressed during the lytic phase of EBV that facilitates viral entry into B cells—the therapy aims to selectively eliminate EBV-infected cells. It is currently undergoing evaluation in Phase 1/2 clinical trials for patients with relapsed or refractory EBV-associated lymphoid neoplasms.
