gp70 CAR T cells are an experimental chimeric antigen receptor (CAR) T cell therapy designed to target the gp70 envelope protein of murine endogenous retroviruses (ERVs). Developed by Genentech, this therapy explores the use of ERVs as universal tumor antigens, which are often highly expressed on the surface of various solid tumor types but absent or low in normal tissues. In preclinical studies using the B16 melanoma model, these CAR T cells demonstrated the ability to suppress tumor growth and recognize tumor cells with high sensitivity, even in MHC-I low environments. The mechanism involves direct tumor cell killing and the local release of interferon-gamma (IFNγ), which upregulates MHC-I on the tumor surface, potentially enhancing endogenous T cell responses.
