The gp75 DNA vaccine is an experimental cancer immunotherapy designed to induce an immune response against melanoma by encoding the melanosomal membrane glycoprotein known as gp75 or tyrosinase-related protein-1 (TRP-1). This antigen is highly expressed in melanoma cells. The mechanism of action involves delivering plasmid DNA encoding the tumor-associated antigen (gp75/TRP-1), leading to *in vivo* expression of the antigen and subsequent activation of both cellular and humoral immune responses targeting melanoma cells. Some versions use self-replicating alphavirus replicons to enhance immunogenicity and break tolerance against this self-antigen[3][2][7]. The primary indication is for melanoma treatment or prevention[1][4].
