Overview

gp96 is a heat shock protein (also known as grp94, endoplasmin, ERp99 or HSP90B1) that functions as an endoplasmic reticulum chaperone involved in the folding and quality control of client proteins such as Toll-like receptors and integrins. It plays a key role in maintaining ER homeostasis and is upregulated under stress conditions. In drug development contexts, "gp96" often refers to therapeutic cancer vaccines that use tumor-derived or recombinant forms of this protein to stimulate immune responses against cancer cells. These vaccines act as immunostimulants by presenting tumor antigens to antigen-presenting cells (APCs), thereby inducing cytotoxic T-cell responses against tumors[3][6]. The most advanced clinical programs involve autologous or allogeneic cell-based vaccines expressing or secreting gp96 for indications such as glioblastoma and other cancers[3][7]. Additionally, peptide antagonists derived from gp96 have shown promise in preclinical models for inflammatory diseases by modulating cytokine production and inflammation[2].
