gp96-Ig is a secreted recombinant form of the heat shock protein gp96 (also known as Grp94 or ERp99), which is naturally found in the endoplasmic reticulum. The secretory form is created by replacing the C-terminal KDEL retention sequence of human gp96 with the hinge region and constant heavy chains (CH2 and CH3) of mouse or human IgG1.\n\ngp96-Ig functions as both an adjuvant and antigen carrier in vaccine formulations. It activates antigen presenting cells (APCs) including dendritic cells, natural killer cells, and cytotoxic T lymphocytes, while efficiently chaperoning peptides for cross-presentation via MHC I to CD8+ T cells. This dual functionality makes it particularly effective at generating robust CD8+ T cell responses at extremely low antigen concentrations.\n\nA key advantage of gp96-Ig vaccines is their ability to induce both systemic immunity (in blood and spleen) and mucosal immunity at potential pathogen entry sites such as the gut, reproductive tract, and lungs. The vaccine approach has been tested in various disease contexts including cancer (particularly non-small cell lung cancer) and infectious diseases like HIV/SIV, malaria, Zika virus, cytomegalovirus, and COVID-19.
