Overview

GPA-TriMAR-T cell is an autologous, genetically engineered T cell therapy designed as a novel immunotherapy for malignant melanoma. Patient-derived (autologous) T cells are isolated and transduced with a lentiviral vector encoding the tri-functional chimeric antigen receptor (TriCAR), known as GPA-TriMAR. This CAR construct features three subunits in its outer membrane domain, which are linked to intracellular signaling domains including 4-1BB and CD3ζ. The engineered cells specifically recognize gp100(209–217) peptides presented by MHC-I on melanoma cells, leading to targeted cytotoxicity against tumor cells expressing this antigen. Additional subunits of the TriCAR are designed to stimulate innate immunity and enhance tumor infiltration by the modified T cells[2][3][4][5][7]. The approach combines elements of both chimeric antigen receptor (CAR) technology and T-cell receptor mimicry.
