GPA33-SADA is an investigational bispecific antibody construct developed using the SADA (Self-Assembling and DisAssembling) platform, designed for pre-targeted radioimmunotherapy of solid tumors that express GPA33, particularly colorectal cancer. The SADA technology enables the antibody constructs to assemble in tetramers to enhance tumor targeting and then disassemble into smaller antibody fragments for rapid renal clearance, thus minimizing systemic exposure. In a pre-targeting strategy, administration of the SADA construct precedes the infusion of a radioactive payload, which subsequently binds selectively to the tumor-localized antibody, delivering targeted radiation to cancer cells. GPA33, the target antigen, is expressed on over 95% of primary and metastatic colorectal cancers. The construct was originally developed at Memorial Sloan Kettering Cancer Center and the Massachusetts Institute of Technology, and is licensed to Y-mAbs Therapeutics for development and commercialization[8][10][12].
