GPA500 is a small-molecule derivative of the natural fungal metabolite galiellalactone, developed by Glactone Pharma as a direct inhibitor of the transcription factor STAT3 for use in oncology and immuno-oncology. GPA500 binds STAT3 intracellularly and blocks its transcriptional activity regardless of upstream activators, suppressing STAT3-driven signaling pathways involved in tumor cell proliferation, survival, drug resistance, and tumor-induced immunosuppression.[1][4][8][9] In preclinical models of prostate cancer, including enzalutamide-resistant disease, GPA500 reduces STAT3 and androgen receptor signaling, downregulates Cyclin D1, and inhibits tumor cell proliferation in vitro and in vivo, supporting its potential as an antineoplastic and immunomodulatory agent and as a lead scaffold for more drug-like STAT3 inhibitors.[1][4][6][8][9]
