GPA7-28z CAR T cells are a type of *chimeric antigen receptor (CAR) T cell therapy* that utilizes a TCR-like CAR construct based on the GPA7 single domain antibody (sdAb) to specifically target the melanoma antigen gp100 presented in the context of HLA-A2, a common human leukocyte antigen. The CAR comprises a GPA7 sdAb fused to a human IgG1 hinge region and transmembrane–intracellular signaling domains from CD28 and CD3ζ. Peripheral blood mononuclear cells (PBMCs) from healthy donors are transduced with a lentiviral vector encoding the GPA7-28z CAR, resulting in T cells that, upon encounter with gp100/HLA-A2 on melanoma cells, expand and exert cytotoxic antitumor activity through mechanisms such as IFN-γ and granzyme B secretion[2]. The therapeutic potential is demonstrated in preclinical models (in vitro and in vivo), showing HLA-A2-restricted, antigen-specific killing of melanoma cells. The GPA7-28z CAR T cells represent a precision immunotherapy approach for HLA-A2 positive melanoma, leveraging a TCR-like recognition for specificity.
