Overview

GPC-1 shRNA is a short hairpin RNA therapeutic candidate designed to silence the expression of Glypican-1 (GPC-1), a cell surface heparan sulfate proteoglycan. GPC-1 is known to mediate cell proliferation, differentiation, and migration, and is frequently overexpressed in various malignancies, including pancreatic, breast, lung, and bone metastatic prostate cancers. In preclinical research, GPC-1 shRNA has been utilized to investigate the protein's role as either an oncogene or a tumor suppressor. While in vitro studies in prostate cancer cell lines (such as PC-3) demonstrated that GPC-1 knockdown reduces cell growth and migration, in vivo mouse xenograft models have suggested a more complex role where GPC-1 inhibition may actually increase tumor size and the expression of metastatic markers like MMP-9 and N-cadherin, potentially due to interactions within the tumor microenvironment.
