GPC2-directed CAR T-cell therapy is an adoptive cell therapy that involves genetically engineering a patient's own T cells to express a chimeric antigen receptor (CAR) specifically designed to recognize and bind to Glypican 2 (GPC2). GPC2 is a protein highly expressed on the surface of various solid tumors, including neuroblastoma, retinoblastoma, medulloblastoma, small cell lung cancer, glioblastoma, and uterine carcinoma, while having minimal expression on normal tissues, making it an attractive target for cancer immunotherapy. Once infused into the patient, these modified CAR T-cells identify and eliminate GPC2-expressing cancer cells. This therapy is being developed by institutions such as the Children's Hospital of Philadelphia, the National Cancer Institute, Tmunity Therapeutics, and Stanford. It is administered intravenously or intracerebroventricularly, depending on the tumor location.
