GPC201 is a combination therapeutic candidate developed by GPCR Therapeutics, consisting of the CXCR4 antagonist plerixafor and the beta-2 adrenergic receptor (ADRB2) antagonist propranolol. The drug is designed based on the discovery that CXCR4 and ADRB2 form heteromers, which play a critical role in the bone marrow niche and tumor microenvironment. By simultaneously inhibiting both receptors, GPC201 aims to enhance the mobilization of hematopoietic stem cells for autologous transplantation in patients with multiple myeloma, a process currently in Phase 2 clinical trials. Beyond stem cell mobilization, GPC201 is being investigated in IND-enabling studies as a chemosensitizer for acute myeloid leukemia (AML) and as an immuno-oncology therapy for solid tumors, where it is intended to disrupt the protective signaling pathways that allow cancer cells to evade treatment.
