GPC3-1XX CAR T cells with IL1RAP CCR are an investigational **IF-BETTER-gated CAR-T-cell therapy** for hepatocellular carcinoma. The engineered T cells use a glypican-3-directed CAR containing the GC33 single-chain variable fragment and a calibrated 1XX signaling module, which preserves GPC3-restricted tumor-cell killing while aiming to improve functional persistence. A separate interleukin-1 receptor accessory protein-directed chimeric costimulatory receptor supplies conditional costimulation after IL1RAP engagement, increasing T-cell proliferation, repeated-stimulation fitness, tumor infiltration, and antitumor activity, particularly against tumors with low GPC3 expression. The construct was reported preclinically by Columbia University investigators.
