Overview

GPC3 CAR-M is a novel cell therapy utilizing chimeric antigen receptor (CAR)-modified macrophages engineered to target Glypican-3 (GPC3), a biomarker for hepatocellular carcinoma (HCC) not expressed in most adult tissues. This therapy involves transducing macrophages with an adenoviral vector delivering an anti-GPC3 CAR construct, which includes an scFv targeting GPC3 and a CD3ζ intracellular domain. These engineered GPC3 CAR-Ms exhibit antigen-specific phagocytosis and tumor cell clearance in vitro and in 3D spheroid models of HCC, alongside promoting the expression of pro-inflammatory (M1) cytokines and chemokines, indicating potent antitumor activity. The drug is being developed by Carisma Therapeutics and Moderna for the treatment of hepatocellular carcinoma.
