GPC3-CAR-Ori2 modified T cells (also known as Ori-C101 or Ori-CAR-001) is an armored, autologous chimeric antigen receptor (CAR) T-cell therapy targeting glypican-3 (GPC3), a cell-surface proteoglycan highly overexpressed in hepatocellular carcinoma (HCC). Developed by OriCell Therapeutics, the therapy is engineered using the proprietary Ori®Armoring platform, which incorporates a novel 'Ori2' element following the 4-1BB and CD3ζ domains. This modification enhances the expansion of memory stem T cells (Tscm), improves in vivo persistence, and helps overcome the immunosuppressive tumor microenvironment of solid tumors. Ori-C101 is administered via intravenous or hepatic arterial infusion and is currently in Phase II clinical development for the treatment of advanced GPC3-positive hepatocellular carcinoma.
