GPC3-CAR-ORI2 T cells are an autologous chimeric antigen receptor (CAR) T cell therapy engineered to target glypican 3 (GPC3), a cell surface protein highly expressed in hepatocellular carcinoma (HCC) and other solid tumors. This "armored" second-generation CAR-T incorporates a proprietary Ori2 element following the 4-1BB and CD3ζ signaling domains, designed to enhance memory stem cell ratio, proliferation, persistence, and antitumor activity compared to traditional CAR-T therapies. Preclinical studies demonstrated improved efficacy both in vitro and in vivo. The product is being evaluated primarily for relapsed or refractory HCC but may have potential for other GPC3-positive solid tumors[1][4].
