**GPC3-CAR-Super IL2** is an experimental, GPC3-directed CAR construct designed for transient **in vivo engineering of macrophages** using lipid nanoparticle-delivered mRNA. The construct combines a glypican-3-targeting chimeric antigen receptor with a Super IL-2 component intended to enhance antitumor immune activation, including CD8-positive T-cell proliferation. It was reported as one component of an LNP-mRNA quadrivalent CAR-macrophage approach administered together with FAP-CAR-ΔTGFβRII in preclinical solid-tumor studies. In an orthotopic mouse hepatocellular carcinoma model, the combined system promoted tumor immune activation, CD8-positive T-cell infiltration and expansion, antigen spreading, immune memory, and complete tumor regression without significant reported toxicity. GPC3-CAR-Super IL2 has not been identified as an independently administered clinical drug product.
