GPC3 CAR-T-AFP-CD3 is an investigational dual-targeting chimeric antigen receptor (CAR)-T cell therapy designed for the treatment of hepatocellular carcinoma (HCC). Developed by OriCell Therapeutics in collaboration with the South China University of Technology and Guangdong Medical University, this therapy utilizes T cells engineered to express a CAR targeting the cell-surface tumor antigen Glypican-3 (GPC3). Furthermore, these cells are modified to secrete a bispecific T-cell engager (BiTE) that targets an HLA-A*02:01-restricted epitope of the intracellular antigen Alpha-fetoprotein (AFP) presented on the tumor surface, while simultaneously engaging bystander T cells via the CD3 receptor. This dual-mechanism approach is intended to enhance anti-tumor activity, improve CAR-T cell persistence, and overcome the challenges of antigen heterogeneity and low expression levels often encountered in liver cancer treatment.
