GPC3-derived peptide refers to a group of investigational peptide-based cancer vaccines, most notably those developed by OncoTherapy Science and Chiba University, designed to induce a cytotoxic T lymphocyte (CTL) response against Glypican-3 (GPC3)-expressing tumors. GPC3 is a cell-surface oncofetal antigen that is highly overexpressed in hepatocellular carcinoma (HCC) and various other solid tumors, including ovarian clear cell carcinoma and gastrointestinal cancers, while remaining minimally expressed in normal adult tissues. The vaccine typically consists of specific HLA-restricted amino acid sequences, such as the HLA-A24-restricted GPC3-144-152 (FVGEFFTDV) or HLA-A2-restricted GPC3-298-306 (EYILSLEEL) peptides. When administered with an adjuvant like Montanide ISA 51, these peptides are presented by MHC class I molecules to activate GPC3-specific CD8+ T cells, which then recognize and lyse GPC3-positive tumor cells. Clinical development has primarily focused on HCC and ovarian clear cell carcinoma, with additional exploratory studies in other solid tumors.
