GPC3-TAC is an autologous T-cell therapy utilizing Triumvira Immunologics' proprietary T-cell Antigen Coupler (TAC) technology. Unlike traditional CAR-T cells that use synthetic signaling domains, the TAC receptor is a hybrid molecule designed to co-opt the natural T-cell receptor (TCR) complex. It consists of an antigen-binding domain targeting Glypican-3 (GPC3), a domain that binds to the CD3 epsilon subunit of the TCR, and a T-cell receptor signaling domain (often derived from CD4). This configuration allows for the recruitment of the full endogenous TCR machinery upon binding to GPC3-expressing tumor cells, potentially leading to more controlled T-cell activation and improved safety and efficacy profiles in solid tumors such as hepatocellular carcinoma.
