GPC3-targeted autologous CAR T cells are a class of genetically engineered cellular immunotherapies designed to target glypican-3 (GPC3), an oncofetal proteoglycan highly expressed in hepatocellular carcinoma (HCC) and other solid tumors, but minimally expressed in normal adult tissues. These therapies involve harvesting a patient's own T cells, genetically modifying them ex vivo to express a chimeric antigen receptor (CAR) specific for GPC3, expanding them, and reinfusing them back into the patient. Various developers, including academic institutions like the Affiliated Drum Tower Hospital of Nanjing University and biopharmaceutical companies like Oricell Therapeutics, AbelZeta, AstraZeneca, and Takeda, are developing GPC3-targeted autologous CAR T-cell therapies (such as Ori-C101, C-CAR031, and TAK-102) for the treatment of advanced hepatocellular carcinoma and other GPC3-positive solid tumors.
