This autologous CAR-T cell therapy, developed by the Chinese PLA General Hospital, targets Glypican-3 (GPC3) for the treatment of advanced hepatocellular carcinoma (HCC). GPC3 is a cell-surface oncofetal antigen that is highly expressed in HCC but has limited expression in normal adult tissues, making it a promising target for cell-based immunotherapy. The therapy involves the genetic modification of a patient's own T cells to express a chimeric antigen receptor (CAR) that specifically recognizes and binds to GPC3-expressing tumor cells, leading to their destruction. A distinguishing feature of this specific program is the use of hepatic arterial infusion for delivery, which aims to increase the local concentration of CAR-T cells within the liver and potentially enhance efficacy while minimizing systemic toxicities. It is currently being evaluated in Phase 1/2 clinical trials, often administered following a lymphodepleting conditioning regimen of fludarabine and cyclophosphamide.
