This investigational *in vivo* chimeric antigen receptor macrophage (CAR-M) therapy is being developed by Carisma Therapeutics in collaboration with Moderna for the treatment of hepatocellular carcinoma (HCC). The therapy utilizes mRNA technology delivered via lipid nanoparticles (LNPs) to genetically reprogram endogenous macrophages and monocytes directly within the patient's body. The mRNA encodes a chimeric antigen receptor (CAR) specific to Glypican-3 (GPC3), a tumor-associated antigen highly expressed in HCC. Once expressed, these CAR-macrophages are designed to recognize and destroy GPC3-positive tumor cells through phagocytosis and the induction of a broader anti-tumor immune response. This off-the-shelf approach aims to circumvent the manufacturing complexities and costs associated with traditional *ex vivo* cell therapies.
