Drug intelligence / Profile preview
GPC3 turbo CAR-T is an investigational, fully human chimeric antigen receptor T-cell (CAR-T) therapy targeting glypican-3 (GPC3), developed by JW Therapeutics for the treatment of advanced GPC3-positive hepatocellular carcinoma (HCC). To overcome the highly immunosuppressive tumor microenvironment (TME) of solid tumors, which typically limits CAR-T cell persistence and induces apoptosis, GPC3 turbo CAR-T is engineered with three distinct armoring elements: a dominant-negative FAS (dnFAS) receptor to block FASL-mediated apoptotic signaling, a dominant-negative TGF-β receptor II (dnTGFβRII) to resist TGF-β-mediated immunosuppression, and an interleukin-7 receptor (IL-7R) to promote T-cell survival and proliferation under cytokine-deprived conditions. In preclinical studies, GPC3 turbo CAR-T cells demonstrated enhanced cytotoxicity, increased proliferation, and superior in vivo tumor clearance and persistence compared to conventional, non-armored GPC3 CAR-T cells.
