GPI-0100

GPI-0100 is a semi-synthetic triterpene glycoside derived from naturally occurring saponins. It functions as an adjuvant in vaccine preparations to enhance the immunogenicity of antigens such as proteins and carbohydrates[1][2][3]. Mechanistically, it acts by upregulating the immune system and stimulating both humoral (antibody-mediated) and cellular (T-cell mediated) immunity, including cytotoxic T lymphocyte (CTL) responses[2][5]. Compared to other saponin-based adjuvants like QS-21, GPI-0100 offers improved stability and a better safety profile[5][6]. The drug has been investigated primarily as an adjuvant for vaccines targeting cancer indications such as breast cancer, kidney cancer (renal cell carcinoma), leukemia (myeloid), melanoma, neuroblastoma, ovarian cancer, prostate cancer[2], and has also been used with viral antigens in preclinical studies. Its development was led by Avantogen and Adjuvantys.
