ConfometRx is developing small molecule agonists of the G protein-coupled receptor 171 (GPR171) for the treatment of chronic and acute pain. GPR171 is a Gi/o-coupled receptor expressed in nociceptive neurons that, when activated by its endogenous ligand BigLEN or synthetic agonists, modulates ion channels to inhibit pain transmission. The program is currently in the preclinical stage, focusing on structure elucidation and lead generation to identify potent and selective candidates. This therapeutic approach aims to provide a novel, non-opioid mechanism for managing inflammatory and neuropathic pain conditions.
