Confo Therapeutics is developing a small molecule antagonist targeting GPR75 (G protein-coupled receptor 75) for the treatment of obesity. GPR75 is a genetically validated target; individuals with loss-of-function variants in the GPR75 gene have been shown to have significantly lower body mass index (BMI) and a reduced risk of obesity. The program leverages Confo's proprietary ConfoBody technology, which uses camelid-derived single-domain antibodies to stabilize GPCRs in specific functional conformations, enabling the discovery of potent and selective small molecules. The program is currently in the lead optimization stage and is being positioned as a next-generation obesity treatment that could potentially be used as a monotherapy or in combination with GLP-1 receptor agonists.
