Overview

GPRC5B siRNA is a small interfering RNA designed to silence the expression of G protein-coupled receptor class C group 5 member B (GPRC5B), also known as retinoic acid-induced gene 2 (RAIG2). GPRC5B is an orphan Class C GPCR that has been implicated in the regulation of calcium signaling within human airway smooth muscle (ASM) cells. Research presented at the ATS 2025 meeting by investigators from the Mayo Clinic demonstrated that silencing GPRC5B with siRNA has an alleviating effect on calcium responses to bronchoconstrictor agonists in ASM cells derived from asthmatic individuals. This suggests that GPRC5B may play a role in the pathophysiology of airway hyperresponsiveness and that its inhibition could represent a novel therapeutic approach for asthma and other obstructive airway diseases.
