Overview

GPRC5D bispecific antibodies represent a class of T-cell redirecting therapies designed to treat multiple myeloma. These biologics simultaneously bind to G protein-coupled receptor family C group 5 member D (GPRC5D), which is highly and specifically expressed on malignant plasma cells, and the CD3 epsilon subunit of the T-cell receptor complex. This dual binding brings T cells into close proximity with myeloma cells, leading to T-cell activation and subsequent lysis of the cancerous cells. Talquetamab (Talvey), developed by Johnson & Johnson, was the first-in-class agent in this category to receive FDA approval. Other investigational agents in this class include Roche's forimtamig (RG6234) and several others in early-stage clinical development, such as LBL-034 and BR109.
