GPRC5D DNA vaccine is a DNA vaccine designed to induce immune responses against G protein-coupled receptor class C group 5 member D (GPRC5D), a protein uniquely expressed in plasma cells and highly overexpressed in multiple myeloma (MM). The vaccine is delivered via intramuscular electroporation and contains plasmid DNA encoding the human GPRC5D antigen; the most advanced constructs use a nanoplasmid vector (Nano-hGPRC5D) for higher gene expression. Preclinical studies demonstrate that vaccination elicits robust humoral and cellular immune responses, suppressing MM tumor growth in mouse models. The vaccine’s mechanistic action is to provoke the immune system to mount targeted antibody and T cell responses against GPRC5D-expressing cells. Combination with PD-1 blockade enhances anti-tumor efficacy, making the vaccine a potential strategy for both prevention and treatment of multiple myeloma, as well as for pre-malignant conditions such as monoclonal gammopathy of undetermined significance (MGUS) and smoldering myeloma[1][2][3][4][5][7].
