GPRC5D-NKG2D BsAb is a bispecific antibody (BsAb) designed for the treatment of multiple myeloma. It is constructed on a human IgG4 platform and features single-chain variable fragments (scFv) that target the orphan G protein-coupled receptor class C group 5 member D (GPRC5D) on myeloma cells and the activating receptor NKG2D on natural killer (NK) cells. Developed by researchers at the City of Hope National Medical Center, the antibody acts as an NK-cell engager, bridging NK cells to tumor cells to facilitate targeted cytotoxicity. In preclinical studies, it has been evaluated both as a purified therapeutic protein and as a secreted component of engineered BCMA-CAR NK cells to address tumor heterogeneity and antigen escape in multiple myeloma models.
