Overview

GPRC5D SIR-T is an investigational cell therapy being developed by Angeles Therapeutics for the treatment of multiple myeloma. It utilizes the company's proprietary Synthetic Immune Receptor (SIR) technology, which involves engineering T-cells to express a chimeric receptor designed to mimic the natural T-cell receptor (TCR) complex. This candidate specifically targets G protein-coupled receptor class C group 5 member D (GPRC5D), an orphan receptor that is highly and selectively expressed on malignant plasma cells in multiple myeloma. By leveraging the cell's endogenous signaling machinery rather than the simplified signaling domains used in traditional CAR-T cells, SIR-T cells aim to achieve superior anti-tumor potency, improved persistence, and a more controlled safety profile. The program is currently in preclinical development.
