Overview

GPRC5DxBCMA SIR-T is a preclinical Synthetic Immune Receptor T-cell (SIR-T) therapy being developed by Angeles Therapeutics for the treatment of multiple myeloma. This dual-targeting cell therapy simultaneously recognizes G protein-coupled receptor class C group 5 member D (GPRC5D) and B-cell maturation antigen (BCMA), two antigens highly expressed on malignant plasma cells. By targeting two distinct markers, the therapy is designed to mitigate the risk of tumor relapse caused by antigen escape, a common limitation of single-target CAR-T therapies. The SIR-T platform utilizes a synthetic receptor architecture that mimics the natural T-cell receptor (TCR) signaling complex, potentially offering more physiological T-cell activation, improved persistence, and better-controlled effector functions compared to traditional chimeric antigen receptor (CAR) designs.
