GPRC5DxNKp30 is a novel bispecific natural killer (NK) cell engager (NKCE) designed for the treatment of multiple myeloma. It simultaneously targets the tumor-associated antigen G protein-coupled receptor family C group 5 member D (GPRC5D) on myeloma cells and the activating receptor NKp30 on NK cells. Developed by researchers at the University Medical Center Schleswig-Holstein, the molecule is an IgG-like construct featuring a Fab fragment derived from the GPRC5D antibody talquetamab and a camelid-derived NKp30 nanobody. By bridging NK cells and tumor cells, it promotes NK cell recruitment, activation, and cytotoxic killing of GPRC5D-expressing malignant cells. Preclinical studies indicate it can effectively lyse myeloma cells, including those with low CD38 expression, and its activity can be further enhanced by an active Fc domain that engages Fcγ receptors to induce phagocytosis.
