GPRP-dextran conjugate is an experimental anticoagulant and antithrombotic agent consisting of the tetrapeptide Gly-Pro-Arg-Pro (GPRP) covalently linked to a dextran polymer. GPRP is a synthetic mimic of the 'knob A' motif (Gly-Pro-Arg) exposed on fibrinogen after thrombin-catalyzed cleavage of fibrinopeptide A. By competitively binding to the complementary 'hole a' on fibrin(ogen) molecules, the conjugate inhibits fibrin polymerization and assembly. The dextran conjugation is designed to prolong the circulation time of the peptide. Preclinical studies conducted at the University of Pennsylvania demonstrate that the conjugate suppresses fibrin formation and alters clot structure without significantly increasing bleeding time, suggesting a potentially safer profile compared to traditional anticoagulants.
