Overview

GPV02 is a glycoengineered protein subunit vaccine currently in preclinical development by GlyProVac. It is designed to target the YghJ antigen, a highly conserved protein found across various *Escherichia coli* strains, including enterotoxigenic *E. coli* (ETEC). The vaccine utilizes GlyProVac's proprietary **BEMAP** (Bacterial Expression of Mammalian Proteins) technology platform, which employs mass spectrometry to identify and replicate O-linked protein glycosylation patterns. By using hyperglycosylated YghJ polypeptides, GPV02 mimics the natural decoration of bacterial proteins to trigger a robust and specific immune response. The candidate is being developed as a multi-indication vaccine for enterotoxigenic *E. coli* infections, diarrhea, urinary tract infections (UTIs), healthcare-associated UTIs, and neonatal sepsis. In 2024, it received funding from CARB-X to support its development as a maternal vaccine aimed at protecting newborns from neonatal sepsis caused by *E. coli*.
