Overview

GPVI-CD39 is a bifunctional recombinant fusion protein being developed by AdvanceCOR for the treatment of acute arterial thrombosis and atherothrombotic conditions. The molecule consists of the extracellular domain of platelet glycoprotein VI (GPVI) linked to the Fc region of human IgG2 and the extracellular domain of the ectonucleotidase CD39 (also known as ADPase or NTPDase1). It operates via a dual mechanism: the GPVI domain targets and binds to collagen exposed at sites of vascular injury, thereby blocking collagen-mediated platelet activation, while the CD39 domain locally degrades adenosine diphosphate (ADP), preventing ADP-induced platelet aggregation and thrombus expansion. This lesion-directed approach is designed to concentrate antithrombotic activity at the site of plaque rupture, potentially reducing the risk of systemic bleeding complications compared to conventional antiplatelet agents.
