Overview

grβ siRNA is a small interfering RNA (siRNA) designed to silence the expression of the glucocorticoid receptor beta (GRβ) isoform. GRβ is an alternative splice variant of the glucocorticoid receptor that lacks the ligand-binding domain and acts as a dominant-negative inhibitor of the transcriptionally active GRα isoform. Elevated levels of GRβ are associated with glucocorticoid insensitivity in inflammatory conditions such as severe asthma. By targeting GRβ mRNA for degradation, this siRNA aims to increase the GRα:GRβ ratio, thereby restoring sensitivity to glucocorticoid therapy and inhibiting pathological processes like airway smooth muscle cell proliferation. Research presented at ATS 2025 has demonstrated that GRβ knockdown significantly reduces mitogen-induced proliferation in primary airway smooth muscle cells, highlighting its potential as a therapeutic approach for steroid-resistant respiratory diseases.
