Overview

GR1014 is a topical formulation of amifostine thiol under development as a **radioprotective** agent to prevent or reduce radiation-induced dermatitis in patients receiving adjuvant radiotherapy for localized breast cancer after lumpectomy.[1][2][11][12][13] It is being evaluated as GR1014 cutaneous gel (GR1014-CG) in phase 2 trials, applied to the breast skin prior to each radiotherapy fraction to mitigate erythema, desquamation, edema, pain, and pruritus without compromising the antitumor efficacy of radiotherapy.[1][2][5][10][11] Amifostine thiol is a free-radical scavenger with well-established radioprotective effects, reducing normal tissue damage from ionizing radiation by donating hydrogen ions to repair DNA and other cellular targets, thereby limiting oxidative injury in irradiated skin.[1][2][11][12]
