Overview

GRAALL-2003 is not a single drug but a multi-agent chemotherapy regimen developed as a pediatric-inspired treatment protocol for adults (ages 15–60) with Philadelphia chromosome-negative acute lymphoblastic leukemia (Ph-negative ALL). The regimen was designed by the Group for Research on Adult Acute Lymphoblastic Leukemia and incorporates intensified doses of nonmyelotoxic drugs commonly used in pediatric protocols, such as prednisone, vincristine, and L-asparaginase. The treatment includes several phases—induction, consolidation blocks with high dose-intensity, delayed intensification, and two years of maintenance therapy. Some adult-specific options like allogeneic stem cell transplantation were retained for high-risk patients. The regimen demonstrated improved complete remission rates and survival outcomes compared to previous adult protocols[1][2][4].
