Overview

Grabody B is a non-neutralizing monoclonal antibody that targets the insulin-like growth factor 1 receptor (IGF1R) and is designed as a molecular shuttle to increase penetration of therapeutics across the blood-brain barrier (BBB)[1][2][5][6][8]. Unlike conventional anti-IGF1R antibodies, Grabody B does not interfere with IGF1R signaling, thereby avoiding disruption of the IGF pathway and associated side effects[1][2]. It is engineered primarily for use as a platform to deliver biologics, including antibodies, polynucleotides, or oligonucleotides (such as siRNA and antisense oligonucleotides), into the central nervous system (CNS), addressing the major obstacle of low brain exposure in CNS drug development[1][2][5][6][7][8]. Preclinical studies and early pipelines leverage its ability to enhance CNS exposure and therapeutic efficacy of fused drugs, particularly in neurodegenerative disorders like Parkinson’s disease[1][2][3][4]. Developed by ABL Bio, Grabody B has been licensed for further neurodegenerative disease research and development by GSK[4][5][6][7][8].
