**Gramicidin S** is a cationic cyclic decapeptide antibiotic produced by *Aneurinibacillus migulanus* and composed of two repeating pentapeptide units. It acts through broad, non-receptor-mediated interaction with microbial phospholipid membranes, causing membrane destabilization, increased permeability, leakage of cellular contents, and microbial cell death. Its membrane activity also creates hemolytic and cytotoxic liabilities that limit use primarily to local topical applications. Gramicidin S is an active ingredient in certain Valenta Pharm Grammidin topical spray products for infectious and inflammatory throat conditions, where it is co-formulated with cetylpyridinium chloride; that combination product should not be conflated with gramicidin S as a single active substance.
