Overview
GRAMOZ-IT is an oral fixed-dose combination therapy developed and marketed by Ingram Biosciences, consisting of the proton pump inhibitor (PPI) rabeprazole and the prokinetic agent itopride. Rabeprazole works by irreversibly inhibiting the gastric H+/K+ ATPase (proton pump) at the secretory surface of parietal cells, thereby suppressing gastric acid secretion. Itopride provides prokinetic activity through a dual mechanism: it acts as a dopamine D2 receptor antagonist and an acetylcholinesterase inhibitor, which increases the concentration of acetylcholine at the neuromuscular junction, enhancing gastrointestinal motility and accelerating gastric emptying. This combination is primarily indicated for the management of acid-related disorders such as gastroesophageal reflux disease (GERD) and peptic ulcers, particularly when these conditions are associated with symptoms of functional dyspepsia or gastroparesis.
