**GRANITE-001** is an individualized, heterologous prime-boost neoantigen cancer vaccine regimen developed by Gritstone bio. It comprises a chimpanzee adenoviral-vector priming vaccine, GRT-C901, followed by a lipid-nanoparticle-formulated self-amplifying mRNA boosting vaccine, GRT-R902; both are customized to encode up to 20 tumor-specific neoantigens identified from an individual patient’s tumor. The regimen is intended to induce neoantigen-specific cytotoxic T-cell responses and tumor-cell killing, and has been studied with immune checkpoint blockade in advanced solid tumors and in frontline maintenance treatment of metastatic microsatellite-stable colorectal cancer.
