Overview

Granulocyte-macrophage colony-stimulating factor plasmid is a plasmid DNA construct encoding the cytokine human granulocyte-macrophage colony-stimulating factor (GM-CSF). It acts as a molecular adjuvant, primarily by enhancing immune responses through induction of dendritic cell proliferation, maturation, and migration, as well as promoting B and T lymphocyte expansion and differentiation. The therapeutic strategy involves using the plasmid as a gene therapy or vaccine adjuvant to increase the immunogenicity of co-administered cancer antigens. GM-CSF DNA has shown potential in early-phase trials as an adjuvant in peptide and DNA vaccines for cancer, notably in melanoma, and has demonstrated a favorable safety profile with primarily mild injection site reactions. It does not function as a standalone cancer therapy but rather augments immune responses to specific antigens when given in combination therapies.
