**Granulocyte-macrophage colony-stimulating factor whole-cell vaccine** is a therapeutic cancer vaccine modality in which tumor cells (often allogeneic or autologous whole cells) are genetically engineered to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF), a cytokine that stimulates myeloid cell development and maturation. The purpose is to enhance antitumor immunity by recruiting, activating, and maturing dendritic cells at the immunization site, thereby promoting the processing and presentation of tumor antigens to T cells. GM-CSF secretion supports the recruitment and activation of neutrophils, monocytes, macrophages, dendritic cells, and leads to enhanced CD4+ and CD8+ T cell antitumor responses. This approach has mostly been developed for immunotherapy of various cancers, including melanoma and prostate cancer, with the intention to create a localized, potent immune response against tumor cells.
