Overview

Grass-SPIRE (also known as ToleroMune Grass) is a synthetic peptide immuno-regulatory epitope (SPIRE) vaccine developed by Circassia (now NIOX Group) for the treatment of grass pollen-induced allergic rhinitis (hay fever). The therapy consists of a mixture of short, synthetic, allergen-derived peptides that represent T-cell epitopes of major grass pollen allergens. These peptides are designed to induce immunological tolerance by targeting T-cells without cross-linking IgE on mast cells or basophils, thereby reducing the risk of systemic allergic reactions during treatment. Despite promising early results, a large Phase 3 trial failed to meet its primary endpoint in 2016, leading to the discontinuation of the program.
