GrB-Fc-4D5 is a preclinical-stage biologic fusion protein being developed by AbBC Therapies for the treatment of HER2/Neu-expressing cancers, including breast, gallbladder, bladder, esophageal/gastric junction, and stomach cancers. The construct comprises a humanized anti-HER2 single-chain variable fragment (scFv) derived from the 4D5 antibody, linked via an IgG-Fc domain to active granzyme B (GrB), a serine protease. This design enables the fusion protein to specifically bind to HER2-positive tumor cells and internalize, delivering granzyme B to the cytoplasm where it triggers apoptosis through multiple pathways. This mechanism mimics natural T-cell and NK-cell mediated cytotoxicity without the requirement for perforin-mediated membrane pore formation.
