GrB-Fc-CEA is a preclinical-stage recombinant fusion protein developed by AbBC Therapies using its proprietary Targeted Granzyme B Immunotherapy (TGI) platform. The therapeutic is designed to target carcinoembryonic antigen (CEA), a tumor-associated antigen overexpressed in various solid tumors, including colorectal, lung, and breast cancers. The drug consists of an active human granzyme B enzyme fused to an antibody targeting CEA via an Fc (immunoglobulin heavy chain) domain. Its mechanism of action involves high-affinity binding to CEA-expressing tumor cells, followed by internalization and the release of granzyme B into the cytoplasm. Once intracellular, granzyme B triggers the pro-apoptotic caspase cascade, inducing programmed cell death. This approach aims to leverage the natural cytotoxic mechanisms of the immune system without the requirement for chemical linkers or synthetic toxins.
